i would say the best thing you could do is make sure your injectors are in tip top shape, do a full tune up with AC Delco tune up parts, run a good synthetic in the engine, trans, and rearend. make sure the cooling system is clean, and keep up on the tire pressure.
Anything beyond that, I would contact CFM-Tech and get their TBI package, comes with a few parts that optimize throttle body/air cleaner setup, and an adjustable fuel pressure regulator.

Thanks guys! My truck is already a TBI, It does need exhaust. I was wondering what size pipe I should use. It has 3 " now but I think that is too big. Any advice on that? The truck does run well. Its not loaded down, but it is a 1 ton.....

If its single 3"' it should be just fine for a 454. if you have duals, i would say 2 1/2" should be more than sufficient. go with a straight through design muffler, the longer the muffler, the mellower the exaust will be.

Gears & Trans type also will affect your mileage.

Yes, this. Also make sure to check the timing with the single tan/black wire disconnected. You should be able to pick up a fair amount of mileage. My '79 1 ton 4x4 crew cab dually even gets 10 mpg.

I've been working on this same thing with my 1 ton TBI 454. So far I've done headers with 2.5" duals, tune up, and little things like tire pressure. My next thing is going to be gears. Mine has 4:11's in it. If yours has 4:11's and your not pulling much you can get away with something a little taller. I'll probably got to a 3:73 and see how that does.
