Does anyone know how much modifications I would need to do I plan on putting it on a k20 frame and axles but as far as transmission goes I plan on using an nv4500 behind a 12v is there a kit that allows me to use this transmission combo and in the end id like to see 600hp

All I can say, is watch the episode of Roadkill where they 12valve Cummins swapped there 74 c20 chevy ramp truck. I learned alot of stuff about how to Cummins swap a square.
